It does so by inhibiting the Cyclooxygenase (COX) pathway, which produces prostaglandins and thromboxanes. Some NSAIDs, especially in high doses, directly deposit in the kidney infrastructure and cause damage, like interstitial nephritis. 
  • Hypertension - In order to increase the blood flow to the kidneys, our body increases blood pressure. Chronic NSAID use can lead to hypertension. As you might know, hypertension is a gateway to many problems including cardiomyopathy and aortic stenosis. 
  • Gastric Ulcers - Prostaglandins are necessary to build a protective barrier on the stomach mucosa to protect against stomach acid. NSAIDs reduce this protective barrier, leading to ulcer formation in the stomach. In severe cases, these ulcers can bleed and become life-threatening.
